Autophagy and Intermittent Fasting: How Many Hours?

Published 22 October 2026 · Last reviewed 22 October 2026 · Figures checked against official sources (how we check)

Short answer: Fasting raises autophagy, the recycling system inside your cells, in many tissues, mostly shown in animal and cell studies. In people the evidence is early. One small study of 18-hour daily fasts found a rise in an autophagy gene marker in blood. No human study shows the hour when autophagy starts.

Autophagy and intermittent fasting are linked mainly through animal and cell research, where going without food raises the cells’ recycling process in many tissues. Human evidence is thin. In one 4-day study, 11 adults ate only between 8 am and 2 pm, and an autophagy gene marker rose in their blood. No human study shows the hour autophagy starts, so treat any exact figure with caution.

What is autophagy?

Autophagy is the process your cells use to break down and recycle their own parts. It clears out damaged cell machinery, misfolded proteins and invading germs. The building blocks it frees up go back into energy and repair, which helps cells cope with stress.

Researchers link autophagy to ageing. When calorie restriction was studied, blocking autophagy weakened its anti-ageing effects. So autophagy may explain part of those effects. Most of this work is in animals and cells.

How does intermittent fasting affect autophagy?

A 2018 review of the research found that fasting and calorie restriction are the strongest non-genetic triggers of autophagy. It rose in many different tissues and organs when food was withheld. Most of that evidence comes from animal and cell studies.

The human evidence is one small study. Eleven adults with overweight ate between 8 am and 2 pm for 4 days, which is an 18-hour daily fast. Compared with eating from 8 am to 8 pm, their morning blood showed more activity of LC3A, a gene used in autophagy. The authors wrote that this schedule “may also increase autophagy” in humans.

The same study found a mixed signal. In the evening, the 8 am to 2 pm schedule raised activity of the MTOR gene. MTOR makes a nutrient sensor that, in lab studies, turns autophagy down. So the evidence in people is early, and the main body of evidence is animal and cell studies.

How many hours of fasting for autophagy?

No human study shows how many hours of fasting it takes. The popular claim that autophagy switches on at 16 hours is not established in people. The same goes for 24 hours, or any other set time.

Claim you may seeWhat the evidence shows
Autophagy starts at 16 hoursNo human study gives a start time.
16:8 is the best fasting time for autophagyNo schedule has been shown to be best. The only human marker study used an 18-hour fast.
Autophagy peaks after 2 or 3 daysNo human study shows the timing, including any peak.
The longer the fast, the more autophagyLonger fasts may support autophagy, but human evidence is limited and no study shows a set number of hours.

The shift towards fat as fuel, with ketones being made, is better measured. In studies it happens somewhere between 12 and 36 hours, depending on your last meal and how active you are. That switch is a change in fuel, and it does not tell you when autophagy begins.

Some people fast past 16 hours hoping to boost autophagy. That is a reasonable experiment if fasting suits you, but there is no proven hour to aim for. The comparison of 16:8, 18:6 and OMAD sets out the common schedules side by side.

What breaks a fast for autophagy?

This site sorts what breaks a fast by four goals: weight loss, blood sugar, autophagy and gut rest. The four goals are our own framework, not a medical standard. Autophagy is the strictest of them, because anything your body senses as food may count.

Protein is the main reason. In cell and animal studies, amino acids such as leucine switch on mTOR, the nutrient sensor that turns autophagy down. Whether a small amount of protein during a fast does this in people is not known. The site’s verdicts are cautious calls based on that lab evidence.

The common “under about 50 calories” idea is a rule of thumb for weight-loss fasts, not an official limit. The autophagy column is stricter, which is why a splash of milk counts. Coffee has its own guide, does coffee break a fast, and evidence that coffee itself raises autophagy comes from mice only. For other drinks, see drinks that keep a fast intact.

Other things linked to autophagy

Eating less overall, without set fasting hours, also raised autophagy in the 2018 review. Exercise raised it in the muscles of mice. Human evidence for both is limited, and neither has a proven dose or timing in people.

You cannot feel autophagy, and it does not show on the scales. The fasting calculator and timer on this site show stages from fed to longer fast. None of them is labelled autophagy, because no one knows when it starts in people.

Is it worth fasting longer for autophagy?

A longer fast for autophagy alone means chasing an effect that cannot yet be timed or measured in people. The measured effects of fasting, such as weight loss, are covered in the guide to fasting’s benefits, each rated by evidence.

Get a doctor’s advice before any fast longer than 24 hours. Once a fast passes 5 days with little or no food, the first meals afterwards carry a risk of refeeding syndrome. Pregnancy, breastfeeding, being under 18 and taking insulin or sulfonylureas all mean no fast without medical advice. Chasing autophagy is no reason to fast after an eating disorder; we advise against that and point you to a doctor.

Frequently asked

At what hour does autophagy start?

No human study shows when it starts. Figures such as 16 or 24 hours rest mostly on animal and cell research. The one human marker study used an 18-hour daily fast for 4 days, and it did not test other lengths.

Does 16:8 fasting cause autophagy?

It may, but no human study has measured autophagy after a 16-hour fast. The closest human evidence used an 18-hour daily fast and found more activity in one autophagy gene in blood cells.

What breaks a fast for autophagy?

On this site, autophagy is the strictest of the four fasting goals. Anything with protein, such as milk, bone broth, collagen or whey, counts as breaking it. Black coffee, black, green or herbal tea, sparkling water and salt do not.

Can you measure autophagy?

Not in everyday life. The human study measured the activity of an autophagy gene in blood cells, which is a marker. It does not count autophagy in your liver or muscles, and nothing you can feel or weigh shows it either.

Does exercise trigger autophagy?

A single bout of exercise raised autophagy in the skeletal and heart muscle of mice. Human evidence is limited, so treat exercise as a possible trigger in people, not a proven one.

Talk to a doctor before fasting longer than 24 hours. If you feel faint, confused or unwell, or have chest pain, stop the fast and get help: call your local emergency number (for example 999 in the UK, 911 in the US and Canada, 112 in the EU). If food, weight or eating is causing you distress, the support services in the footer can help.
